Complementary PCAF–Coenzyme A Mutagenesis: Chemoenzymatic Synthesis of a Novel Enlarged Coenzyme A Analogue and Evaluation of Its Biological Activity

Written by Leang Khim on October 18, 2010 – 5:00 am -

Sampling substrate specificity: An enlarged coenzyme A analogue, acetyl-3′′-O-benzyl-CoA, has been synthesised from D-pantethine by using the enzymes DPCK and PPAT. The analogue was evaluated to determine if it is a selective cosubstrate for the histone acetyltransferase PCAF mutant Q581A so as to allow it to be used to identify the non-histone substrates of this enzyme.
